There are regular buses to Kos Town from the airport which go via Mastihari.

In summer 2013 there were al least 8 buses per day (Mon-Sat) from the airport to Kos Town and 5 on Sundays. Times for some reason vary quite a bit from day to day, possibly to account for aircraft arrivals. There's a full timetable at www.mastihari-uncovered.com
EDIT; just checked and the timetable has now reverted to winter schedules so just as a guide, buses are at 0755, 0955, 1020/1035, 1115/1135, 1150/1225/1345, 1530/1615, 1650/1720/1725, 1825, and 1940. bookmark thie link and start to check in May when the 2014 schedules will be on showEdited: 28 November 2013, 00:12

Snakebite we used resorthoppa this October to take us to and from Kos town lambi and the full amount for two of us was £18 in a small mini bus it was a perfect way to get to your hotel and be picked up from
