Barring an accident that closes the roadway, the drivetime is between 4-5 hours, depending on stops.
It's a lovely drive, just obey the speed limit and enjoy the view.
ROUGHLY - Its about 168 miles from MIA to Key West.

15 minutes to get back to US 1 in Key Largo.
15 minutes to drive through Key Largo.
20 minutes to get to Lor-E-Lei
45 minutes to park, grab another cold on, use the facilities, and get back on the road.

40 minutes to have a cold one at Sparky's Landing and say high to barb the bartender from buffettguy.
Another hour to Key West.
This is just a guess, as I haven't done this yet (this year).
Gosh, I'd love to have some fellow Iowans to help us along the way, but we rented a wee little convertible, so there's probably not room for our luggage and capable co-pilots!
Maybe the next time!
I suppose that now that you've used ol' buffettguy for all his info, you'll just toss this Minnesotan to the side.
I made that drive in July, it was great. Just be sure your suitcase is not too large for the little car. The only way we could get ours in was to let the top down and put it in the backseat. Which is not a problem, unless it needs to be done during a heavy rain. It was soooooo much fun. The drive is great. Hope you have a wonderful trip.
You've read this, right?
The lovely Mrs. Gam! Welcome......
there is a mile by mile guide in addition to the one linked above, it's a biggun, 28 pages if you care to print it.
Its never taken me more than 3 hrs to drive from Mia to KWF. It always takes a lot longer to get out. My wife won't let me drive home without stopping at at least 3 tourist shops.
Avoid peak times, and don't think youre going to leave on Sunday morning and beat the traffic.
