MONA has its own bus that runs from the Brooke Street Pier on the docks directly to MONA: https://mona.net.au/visit That's what i would use. :)

You could catch a 520, 521 0r 522 from Elizabeth Street in the city centre. They run along Main Road about 300 metres from MONA. They're all direct buses. Map of route here: http://metrotas.com.au.s3.amazonaws.com/wp-content/uploads/2015/12/15542_MTAS_network_guide_north-web.pdf

You can pay cash on the bus or buy a Greencard at the Metro shop at 22 Elizabeth Street. Fares explained here: http://www.metrotas.com.au/fares/urban-fares/

Alternative to bus is the ferry at $22 return (same price a MONA bus) - interesting trip and different approach to the museum but with 100 steps up to the entrance (there is a video installation at the wharf when you arrive).
