Yes, they do. I left my luggage while I went sightseeing on the day I checked out. They give you luggage tags to pick up your luggage when you're ready to depart.

As I recall, there is an escalator going up to ground level, no stairs. There is a luggage turnstile too.

As I recall, there is an escalator going up to ground level, no stairs. There is a luggage turnstile too.
Maybe consider the Heathrow Express to Paddington and then a black cab to the hotel? It would cost more, but with luggage and children, it might make your life easier and less stressful. Heathrow Express is free for children under 15 with a paying adult. The only challenge might be if you miss the last Healthrow express.
The Tube is usually very crowded from Heathrow. You are going really late at night, but I haven't been able to sit down the past two times I've taken the Tube from Heathrow.
