 Assisi from Rome or Siena? 

First time to Italy for two weeks, and basing out of Rome, and Siena. Which will be easiest transit to Assisi?

Also, although not Catholic, want trip to be of a personal, and spiritual nature. Any recommendations on private guide, either local or out of the base city?

Siena is closer as the crow flies, but there are no direct connections; from Rome you can take one of the regional trains to Perugia, they stop all in Assisi.

Definitely Rome would be your easiest route to take by train or car,
