Sure but it will cost money. You'll have to check with the airline to find out how much.
Check carefully that by cancelling one portion of your ticket, the return isn't automatically cancelled.
I bought an airline ticket months ago that flies from Korea to USA to Canada. I'm thinking of taking a small one week vacation in the US when I land in the US. Is it possible to cancel the USA to Canada flight and fly to a different part of the US then fly back to Canada?

I'm not sure you stated your question correctly.
Is your flight really only Korea to USA to Canada. And thats it? You stay in Canada afterwards?
If it is, then yes you can collect your luggage and just walk away after you go through customs in the US.
If you are going somewhere else after Canada on the same tickets, then the answer is no you cant do that without amending your ticket which you may or may not be allowed to do.
